Title of Invention | A PROCESS FOR INHIBITING THE ADENOSINETRIPHOSPHATASE (ATPASE) ACTIVITY OF FISH ACTOMYOSIN USING HEAVY METALS |

Abstract | The present investigation provides a process for inhibiting the adenosinetriphosphatase (ATPase) activity of fish actomyosin using heavy metals, it relates to the inhibition of nucleotide degrading enzyme adenosinetriphosphatase (ATPase) by zinc chloride. The process steps are: i) isolating the actomyosin from Indian oil sardine (Sardinella longiceps), ii) estimating the Ca2+ ATPase activity of actomyosin in presence of zinc chloride at various pH values and temperaturesjii) elucidating the inhibitory capacity of these compounds on Ca2+ ATPase activity at varied pH conditions and temperatures, which illustrates that the inhibition is effective in the pH range of 7.0 to 9.0 and the temperature range of 25°C to 45°C. |
Full Text | We claim: 1. A process for inhibiting the adenosinetriphosphatase (ATPase) activity of fish actomyosin using heavy metals comprising; a) isolating actomyosin from fish; b) quantifying the enzyme activity of actomyosin by known methods; c) estimating the ATPase activity of actomyosin in presence of selected zinc compounds in the concentration range of 1x10-5M to 1x10-2M in reaction mixture; d) elucidating the inhibitory effect of above compounds at pH in the range of 7-9 and temperature at the range of 25°C- 45°C; 2. A process as claimed in claim 1, wherein the additive used is ZnCl2 in the range of 1x10-3M to 1x10-2M. 3. A process as claimed in claims 1&2, wherein the fish actomyosin used is Indian oil sardine (Sardinella longiceps) actomyosin. 4. A process for inhibiting the adenosinetriphosphatase (ATPase) activity of fish actomyosin using heavy metals substantially as herein described with reference to the examples accompanying this specification. Reference may be made to US patent no.6573061 .Hartman et a/., June 3 2003 sarcomere protein composition where describes about the compounds that modulates the biological activity of biochemically functional sarcomere. Objectives of invention The main objective of the invention is to provide a process for inhibiting the adenosinethphosphatase (ATPase) activity of fish actomyosin using heavy metals. Summary of invention Accordingly the present invention relates to a process for inhibiting the adenosinetriphosphatase (ATPase) activity of fish actomyosin using heavy metals comprising; a) isolating actomyosin from fish; b) quantifying the enzyme activity of actomyosin by known methods, c) estimating the ATPase activity of actomyosin in presence of selected zinc compounds in the concentration range of 1x10-5M to 1x10-2M in reaction mixture; d) elucidating the inhibitory effect of above compounds at pH in the range of 7-9 and temperature at the range of 25°C- 45°C; In an embodiment of the process the additive used is ZnCI2 in the range of 1x10-3M to 1x10-2M. |
